The 72-year-old Copacabana singer had just finished a sold-out concert in Memphis when he had to be rushed back to California, according to a statement on his Facebook page.
Manilow had surgery on Monday but continued with his One Last Time - One Last Tour and had scheduled stops in Highland Heights in Kentucky on Thursday and Nashville on Friday.
Legendary singer-songwriter Barry Manilow (pictured here performing last week) has been rushed to a Los Angeles hospital following complications from emergency oral surgery he had earlier this week
The dates are being re-scheduled and will be announced shortly, according to the statement.
It is unclear is Manilow will be able to attend the Grammy Awards on Monday, where he is nominated for his latest album My Dream Duets.
Victoria Varela, Manilow's publicist, said the the singer is still 'perfectly healthy' and is no longer in the hospital.
'Other than the dental issue, he is doing great,' she told CNN.
Manilow announced last year he would be embarking on his last big tour.
'It doesn't mean I'm retiring or anything,' he told Billboard. 'I'll do shows and I'll promote albums if I make any more, but no more big tours.'
'It's too much packing. It's 40 years - more than that, really - of packing and waiting for room service.'
'People think it's glamorous, but glamorous is the last word I would use for this job... I haven't gone sightseeing, ever. It's a job.'
Manilow told Billboard the show would be one of the longest he has done 'for awhile' and filled with as many of his well-known songs as possible.
'I'm trying to get in as many of the big hits as I can,' he said. 'Hopefully people will remember them, but I'm going to do them anyway.
Manilow had his first number one hit, Mandy, forty years ago and had a string of hits in the 1970s and 1980s, including Can't Smile Without You and Could It Be Magic.
He released two albums in 2014 and married his long-time manager Garry Kief in a private ceremony at their Palm Springs home.
The nuptials were so secret that many of Manilow's friends didn't even find out until their first wedding anniversary.
Manilow, 71, and Kief, 66, married at their 64-acre compound in Palm Springs, California in April last year, but news of the nuptials only broke this week as they close in on their first anniversary.
